JOE is carboxy-4',5'-dichloro-2',7'-dimethoxyfluorescein, which has two chlorine and two methoxy substituents, and is a kind of fluorescein derivative. Dyes containing JOE groups are a class of xanthene dyes that emit in the yellow range of the spectrum and can be used as a substitute for HEX dyes.

Background

Fluorescent probes have the advantages of convenience, good selectivity, and high sensitivity, and have been widely used in analytical chemistry, life sciences, clinical medicine, pharmacy and other fields. Fluorophore is the most basic component of fluorescent probe, which could identify the object to be detected and express it as corresponding spectral information. Xanthene dyes represented by fluorescein and rhodamine have a large conjugate plane and a good rigid structure, and more widely used than any other fluorophores. Compared with other fluorophores, xanthene dyes have impressive photophysical properties, such as large extinction coefficient, high fluorescence quantum yield, longer absorption and emission wavelengths in the visible light region, not easy to photobleach, and abundant chemical modification sites.

JOE, as a chlorinated fluorescein, belongs to the xanthene type dye, with high fluorescence quantum yield and weak pH sensitivity. At pH=9, JOE's absorption wavelength is 520 nm, emission wavelength is 548 nm, emission is in the yellow range, and the spectrum matches the channel of HEX dye.

Application of JOE

Dye containing JOE group is often used for automatic DNA sequencing, labeling dATP or ddATP, and is also often used for electrophoresis detection and PCR product quantification. Dyes containing JOE groups are also used to label nucleotides. JOE dyes containing alkynyl or azido moieties can be used for click chemistry reactions with azides or alkynes. JOE phosphoramidite dyes can be used in the synthesis of labeled oligonucleotides.
