Cyanine7.5 dyes are a class of near-infrared (NIR) fluorescent cyanine fluorescent dyes with poor water solubility, high extinction coefficient and bright fluorescence. These dyes are suitable for pH insensitive and have long emission peaks greater than 800 nm. They have high specificity and sensitivity, and the fluorescence quantum efficiency is lower than other dyes, generally 0.1-0.2. Dyes with similar fluorescent properties include Alexa Fluor 790, DyLight 800, indocyanine green (ICG) and so on.
Characteristics of Cyanine7.5 Dyes
Ordinary Cyanine7.5 dyes have poor water solubility, so the organic co-solvent such as DMF, DMSO and acetonitrile, etc. are required for the aqueous labeling reaction system. Light waves have excellent tissue penetration in the near-infrared region, which are especially suitable for in vivo imaging of live small animals to replace radioactive elements.
Application of Cyanine7.5 Dyes
Near-infrared radiation of Cyanine7.5 dyes is commonly used to label small animals for in vivo imaging. Because dyes penetrate tissue readily, so that they can be used to label various biomolecules such as amine-containing proteins, peptides and antibodies, etc. to track their distribution in vivo. But for biomolecular labeling, the water solubility of labeling reagents is low, and organic co-solvents must be used to dissolve the molecules for effective reactions. In addition, due to the low near-infrared fluorescence background of labeled secondary antibody, they are often used in Western Blotting. Some cyanine 7 fluorophores are derived from the well-known clinical dye Indocyanine green (ICG) and have conventional ICG fluorescent cores. There are also some commercial cyanine7.5 dyes that are cyclohexene-cured, and have similar fluorescent properties to ICG-like cyanine7.5 dyes.
