Product Introduction
Sulfo-Cyanine5 dUTP (deoxyuridine triphosphate) is a modified nucleoside triphosphate for the enzymatic labeling of DNA. The triphosphate is an analog of Cy5-dUTP. However, unlike aminoallyl dUTP directly modified with dye, this triphosphate contains a longer linker arm that increases the efficiency of its incorporation by usual polymerases, like Taq polymerase.Sulfo-Cyanine5 is a bright far red emitting dye for Cy5 channel.

Fluorescent Labeling of DNA for Visualization and Quantification: Sulfo-Cyanine5 dUTP is widely used for the fluorescent labeling of DNA, enabling researchers to visualize and quantify DNA in various biological samples. Its incorporation into DNA through enzymatic reactions, such as PCR or nick translation, allows for the generation of fluorescently labeled DNA probes. These probes are instrumental in techniques like fluorescence in situ hybridization (FISH) and flow cytometry, where they facilitate the identification and quantification of specific DNA sequences or chromosomal regions. This application is crucial for studying gene expression, genetic mapping, and diagnosing chromosomal abnormalities in both clinical and research settings.
Real-Time PCR and qPCR Applications: Sulfo-Cyanine5 dUTP can be incorporated into DNA during PCR and quantitative PCR (qPCR) reactions to provide a non-radioactive, fluorescence-based method for detecting and quantifying DNA. The fluorescence emitted by the incorporated Sulfo-Cyanine5 dUTP is directly proportional to the amount of DNA synthesized, making it an excellent tool for real-time monitoring of DNA amplification. This approach enhances the sensitivity and specificity of qPCR assays, particularly in multiplex assays where multiple targets are detected simultaneously. Its application in qPCR is especially valuable for gene expression analysis, pathogen detection, and genetic variation studies.
DNA Damage and Repair Studies: Incorporation of Sulfo-Cyanine5 dUTP into DNA is a powerful method for studying DNA damage and repair mechanisms. When cells are exposed to DNA-damaging agents, the presence of Sulfo-Cyanine5 dUTP can be used to label and detect sites of DNA synthesis during repair processes. This application is crucial for understanding the dynamics of DNA repair pathways, such as nucleotide excision repair and homologous recombination. By monitoring the incorporation of Sulfo-Cyanine5 dUTP, researchers can gain insights into the efficiency and fidelity of DNA repair mechanisms, which has significant implications in cancer research and the development of therapies targeting DNA repair pathways.
High-Resolution Imaging of DNA Dynamics in Live Cells: Sulfo-Cyanine5 dUTP is employed in live-cell imaging studies to track DNA dynamics and chromatin organization in real time. Its incorporation into newly synthesized DNA allows researchers to visualize and analyze DNA replication, cell cycle progression, and chromatin architecture within living cells using advanced fluorescence microscopy techniques. This capability is essential for studying cellular processes such as mitosis, DNA replication timing, and chromatin remodeling. The high-resolution imaging enabled by Sulfo-Cyanine5 dUTP provides valuable information about the spatial and temporal organization of the genome, contributing to a deeper understanding of cellular function and gene regulation.
