Product Introduction
ATTO 633 belongs to a new generation of fluorescent labels for the red spectral region. The dye is designed for application in the area of life science, e.g. labelling of DNA, RNA or proteins. Characteristic features of the label are strong absorption, high fluorescence quantum yield, high photostability, good water solubility, and very little triplet formation. ATTO 633 is a cationic dye. After coupling to a substrate the dye carries a net electrical charge of +1. In common with most ATTO-labels, absorption and fluorescence are independent of pH, at least in the range of pH 2 to 11, used in typical applications.ATTO-Dye Labeled Phospholipids Sigma-Aldrich offers a variety of glycero-phospholipids carrying one or two fatty acid groups (lipophilic groups) and a phosphate ester residue (hydrophilic group). They are labeled at the hydrophilic head group. After incorporation of the phospholipid into a membrane the fluorophore is located at the water/lipid interface of the membrane. We currently provide 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE), 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), palmitoyl-sn-glycero-phosphoethanolamine (PPE), and 1,2-dimyristoyl-sn-glycero-3-phospho-ethanolamine (DMPE) labeled with ATTO-dyes.find more information here

Application
ATTO 633 PPE is a far-red ATTO 633-based fluorescent dye reagent designed for incorporation into fluorescence labeling workflows through a functional "PPE" handle that supports downstream conjugation and probe construction. Its strong red-shifted emission is commonly leveraged for multiplex-friendly imaging and for minimizing spectral cross-talk with green/orange channels in microscopy and flow cytometry. Researchers use ATTO 633 PPE to build fluorescent conjugates, trace biomolecules, and generate labeling standards for quantitative fluorescence readouts.
1. Fluorescent Conjugate Labeling
ATTO 633 PPE is used by chemical biology and biomaterials teams to prepare fluorescent conjugates for tracking proteins, peptides, and other biomolecular building blocks in solution or on surfaces. In labeling workflows, the ATTO 633 emission is frequently selected to support multicolor experiments where separation from shorter-wavelength dyes is important. The PPE functional handle enables researchers to incorporate the dye into their conjugate design so that the resulting labeled construct can be imaged, quantified, or used as a fluorescence reagent in assay development.
2. Fluorescence Microscopy Imaging
ATTO 633 PPE is applied in fluorescence microscopy to visualize labeled biomolecules and to support multichannel cellular imaging experiments where far-red detection improves channel separation. Imaging groups commonly use ATTO 633-based reagents to reduce interference from autofluorescence and to maintain clear signal readouts in complex samples such as labeled cell preparations and biomaterial-associated systems. Labeled conjugates produced from ATTO 633 PPE are often used to study localization patterns, binding/association behavior, and spatial distribution in microscopy-based workflows.
3. Flow Cytometry Fluorescent Tagging
ATTO 633 PPE is frequently incorporated into fluorescent tagging strategies for flow cytometry, enabling far-red detection of labeled targets in single-cell analysis. Assay developers and cytometry users value ATTO 633 emission for multiplex panels because it can be assigned to channels that are spectrally separated from common green/orange fluorophores. Conjugates prepared using ATTO 633 PPE are used to quantify fluorescence intensity distributions, compare labeling conditions, and support gating strategies in fluorescence-based cytometry experiments.
4. Molecular Imaging Reagent Development
ATTO 633 PPE supports molecular imaging reagent development in research settings where far-red fluorescence is preferred for clearer visualization and robust detection in labeled constructs. Probe and platform developers use ATTO 633 PPE to generate dye-bearing intermediates that can be incorporated into larger imaging tools, including fluorescent tracers and labeled affinity reagents used in imaging studies. The reagent format is commonly selected when the downstream application requires a defined ATTO 633 fluorophore presence while leaving flexibility for conjugate assembly into the final imaging reagent.
