Product Introduction
ATTO 590 is a novel fluorescent label belonging to the class of Rhodamine dyes. The dye is designed for application in the area of life science, e.g. labeling of DNA, RNA or proteins. Characteristic features of the label are strong absorption, high fluorescence quantum yield, high thermal and photo-stability, and very little triplet formation. The dye is highly suitable for single-molecule detection applications and high-resolution microscopy. The alkyne modification is used in the Huisgen reaction (“Click Chemistry“).find more information here

Application
ATTO 590 alkyne is an ATTO 590 fluorophore functionalized with an alkyne handle, enabling copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) for fluorescent labeling and probe construction. Its bright red/orange emission makes it well suited for fluorescence microscopy, imaging-based assays, and multicolor workflows where an alkyne-bearing dye conjugate is required.
1. Biomolecule Click Labeling
ATTO 590 alkyne is used in bioconjugation workflows to introduce a red-emitting fluorescent tag onto azide-functional biomolecules, including peptides, proteins, and affinity ligands. Researchers commonly employ it to generate fluorescent conjugates for binding studies, tracking experiments, and reagent development when the labeling strategy benefits from a defined alkyne handle and a robust click coupling step. The dye's compact labeling format supports downstream purification and conjugate characterization for microscopy and fluorescence-based readouts.
2. Fluorescence Microscopy Tracing
ATTO 590 alkyne is frequently incorporated into fluorescent labeling schemes for cellular and biomaterial imaging, where an azide-functional target is tagged with a red fluorophore for visualization. In microscopy workflows, the alkyne functionality allows the dye to be installed onto azide-bearing scaffolds, linkers, or biomolecule coatings, enabling spatial tracking of labeled components in fixed-cell experiments and imaging studies of labeled constructs. The resulting ATTO 590-labeled conjugates are used to follow localization, uptake-like behavior in model systems, and distribution patterns in fluorescence imaging experiments.
3. Multicolor Flow Cytometry Dyes
ATTO 590 alkyne is applied as a click-compatible fluorescent reagent to prepare red-channel labeling reagents for flow cytometry panels built around azide-bearing targeting molecules. By coupling the dye to azide-functional antibodies, lectins, or other affinity reagents, researchers can generate consistent fluorescent conjugates for cell population analysis and gating workflows that require a defined spectral position. The alkyne handle supports modular panel construction, allowing researchers to swap or standardize labeling components while keeping the fluorophore identity consistent across experiments.
4. Surface And Polymer Functionalization
ATTO 590 alkyne is used to fluorescently functionalize azide-containing surfaces, coatings, and polymeric materials for materials characterization and imaging of engineered interfaces. In biomaterials research, the dye is installed onto azide-functional hydrogels, beads, or surface-modified substrates to visualize attachment, coverage, and spatial patterning. This approach supports fluorescence-based quality control and localization studies in which the fluorescent signal originates from a stable, click-derived dye conjugation to the material scaffold.
