Product Introduction
ATTO 550 is a fluorescent label related to the well-known dye Rhodamine 6G. The dye is designed for application in the area of life science, e.g. labeling of DNA, RNA or proteins. Characteristic features of the label are strong absorption, high fluorescence quantum yield, high thermal and photo-stability, and very little triplet formation. The dye is highly suitable for single-molecule detection applications and high-resolution microscopy.ATTO 550 is a cationic dye (charge +1). As supplied ATTO 550 consists of three isomers with identical absorption and fluorescence.ATTO-Dye Labeled Phospholipids Sigma-Aldrich offers a variety of glycero-phospholipids carrying one or two fatty acid groups (lipophilic groups) and a phosphate ester residue (hydrophilic group). They are labeled at the hydrophilic head group. After incorporation of the phospholipid into a membrane the fluorophore is located at the water/lipid interface of the membrane. We currently provide 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE), 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), palmitoyl-sn-glycero-phosphoethanolamine (PPE), and 1,2-dimyristoyl-sn-glycero-3-phospho-ethanolamine (DMPE) labeled with ATTO-dyes.find more information here

Application
ATTO 550 PPE is a fluorescent labeling reagent built around the ATTO 550 dye, designed to support robust conjugation workflows while maintaining strong red-orange fluorescence for imaging and assay development. Its reactive functionality is intended for attaching the fluorophore to biomolecules or materials, enabling traceable targets in fluorescence microscopy, flow-based readouts, and fluorescence-based analytical assays. Researchers commonly use ATTO 550 PPE when a bright, spectrally separated red-emitting fluorophore is needed for multicolor experiments and quantitative fluorescence measurements.
1. Fluorescent Protein Labeling
ATTO 550 PPE is used in protein labeling workflows where red fluorescence readouts are required for tracking biomolecules, monitoring conjugate formation, or visualizing labeled targets in cellular and biochemical assays. In research labs, it is frequently applied to generate fluorescent protein conjugates for microscopy studies, fluorescence-based binding experiments, and reagent development for downstream assays. The ATTO 550 emission profile supports multicolor panels by providing spectral separation from common green and yellow dyes, making it a practical choice for multiplex imaging and comparative quantification.
2. Flow Cytometry Staining
ATTO 550 PPE supports fluorescence staining strategies for flow cytometry experiments that require a red-emitting reporter to quantify labeled cell-associated components. Typical use cases include labeling antibodies, affinity reagents, or other targeting ligands to produce ATTO 550-tagged conjugates used in cell-surface or intracellular staining workflows. The dye's compatibility with standard fluorescence instrumentation enables researchers to incorporate ATTO 550 signals into multichannel gating schemes for population-level analysis of labeled biomolecules.
3. Fluorescence Microscopy Imaging
ATTO 550 PPE is applied to fluorescence microscopy for visualizing labeled biomolecules and tracking spatial distribution in fixed-cell or prepared sample formats. Researchers use ATTO 550-conjugated reagents to map binding patterns, follow biomolecular localization, and generate fluorescent imaging tools for mechanistic cell biology studies. The red-orange emission of ATTO 550 helps reduce spectral overlap with many common fluorophores, supporting clearer channel separation in multicolor imaging experiments.
4. Fluorescent Bioassay Development
ATTO 550 PPE is commonly used during fluorescence assay development to prepare fluorescent conjugates that serve as reporters in binding, detection, and quantification formats. In assay development workflows, ATTO 550-labeled biomolecules can be incorporated into homogeneous or wash-based fluorescence readouts to monitor assay progress and reagent performance. The dye's strong fluorescence supports the construction of calibration standards and labeled reagents for reproducible fluorescence measurements across experimental batches.
