Product Introduction
ATTO 532 is a fluorescent label related to the well-known dye Rhodamine 6G. Characteristic features of the label are strong absorption, high fluorescence quantum yield, high photostability, and excellent water solubility. Thus ATTO 532 is highly suitable for single-molecule detection applications and high-resolution microscopy such as PALM, dSTORM, STED etc. Additionally the dye highly qualifies to be applied in flow cytometry (FACS), fluorescence in-situ hybridization (FISH) and many more. The fluorescence is excited most efficiently in the range 515-545 nm.A suitable excitation source for ATTO 532 is the 532 nm output of the frequency-doubled Nd:YAG laser.ATTO-Dye Labeled PhospholipidsSigma-Aldrich offers a variety of glycero-phospholipids carrying one or two fatty acid groups (lipophilic groups) and a phosphate ester residue (hydrophilic group). They are labeled at the hydrophilic head group. After incorporation of the phospholipid into a membrane the fluorophore is located at the water/lipid interface of the membrane. We currently provide 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE), 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), palmitoyl-sn-glycero-phosphoethanolamine (PPE), and 1,2-dimyristoyl-sn-glycero-3-phospho-ethanolamine (DMPE) labeled with ATTO-dyes.find more information here

Application
ATTO 532 PPE is a reactive fluorescent dye building block designed for labeling workflows where an ATTO 532 fluorophore is coupled into polymer, biomolecule, or surface formats using a PPE-type reactive handle. Its strong green-to-yellow emission is commonly leveraged in fluorescence microscopy and fluorescence-based assays, particularly when robust covalent attachment is needed to minimize dye loss during washing and analysis. Researchers use ATTO 532 PPE to prepare conjugates and labeled materials for imaging, quantitative fluorescence readouts, and multicolor experimental design where ATTO 532 spectral positioning is advantageous.
1. Fluorescence Microscopy Labeling
ATTO 532 PPE is frequently used by cell biology and materials imaging groups to generate fluorescent labeling reagents for microscopy, including covalently stained polymers, bead-based tracers, and labeled biomolecule conjugates that require stable retention during fixation or multi-step staining. In practical imaging workflows, the dye's ATTO 532 emission is selected for bright visualization of labeled targets in fluorescence channels that match common microscope filter sets. Because the reagent is intended for downstream conjugation into defined constructs, it is often incorporated into labeling strategies for tracking distribution, comparing uptake or binding outcomes, and visualizing surface-associated or matrix-embedded materials.
2. Flow Cytometry Fluorescent Conjugates
ATTO 532 PPE is used in flow cytometry workflows where researchers need a fluorescent tag that can be covalently introduced into antibodies, affinity reagents, or other labeling carriers to support consistent signal across wash steps. By converting ATTO 532 PPE into a conjugate format compatible with labeling protocols, laboratories can generate cell-associated fluorescence for population analysis, gating-based quantification, and assay development using ATTO 532 as the reporter channel. This reagent format is particularly relevant when conjugate stability is important for reproducible staining intensity across replicates and experimental runs.
3. Biomaterial Surface Functionalization
ATTO 532 PPE is applied in biomaterials research to fluorescently functionalize surfaces and bulk materials, enabling visualization of coating uniformity, surface coverage, and material localization in imaging studies. Materials scientists often incorporate the reagent into polymer layers, hydrogel formulations, or surface-modified substrates to create stable fluorescent reporters that remain associated with the material during rinsing, incubation, and time-course experiments. The resulting labeled surfaces are then used to study material-cell interactions, diffusion or retention of bound components, and spatial organization within engineered matrices.
4. Fluorescence Bioassay Development
ATTO 532 PPE supports fluorescence assay development in chemical biology and assay engineering settings by providing a dye source for constructing fluorescent conjugates used as readout reagents. Assay developers use ATTO 532-labeled constructs to create labeling-based fluorescence assays, including reagent systems where covalent dye attachment helps reduce background from dye leaching. In these workflows, ATTO 532 fluorescence serves as a measurable signal for endpoint quantification and method optimization, including reagent screening and protocol tuning where consistent labeling chemistry and stable dye retention are important for reliable plate-based measurements.
