Product Introduction
ATTO 532 is a fluorescent label related to the well-known dye Rhodamine 6G. Characteristic features of the label are strong absorption, high fluorescence quantum yield, high photostability, and excellent water solubility. Thus ATTO 532 is highly suitable for single-molecule detection applications and high-resolution microscopy such as PALM, dSTORM, STED etc. Additionally the dye highly qualifies to be applied in flow cytometry (FACS), fluorescence in-situ hybridization (FISH) and many more. The fluorescence is excited most efficiently in the range 515-545 nm.A suitable excitation source for ATTO 532 is the 532 nm output of the frequency-doubled Nd:YAG laser.ATTO-Dye Labeled PhospholipidsSigma-Aldrich offers a variety of glycero-phospholipids carrying one or two fatty acid groups (lipophilic groups) and a phosphate ester residue (hydrophilic group). They are labeled at the hydrophilic head group. After incorporation of the phospholipid into a membrane the fluorophore is located at the water/lipid interface of the membrane. We currently provide 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE), 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), palmitoyl-sn-glycero-phosphoethanolamine (PPE), and 1,2-dimyristoyl-sn-glycero-3-phospho-ethanolamine (DMPE) labeled with ATTO-dyes.find more information here

Application
ATTO 532 DMPE is a fluorescent dye built on an ATTO 532 core, formulated for membrane-associated labeling through its DMPE (1,2-dimyristoyl-sn-glycero-3-phosphoethanolamine) lipid functionality. In fluorescence workflows, it is commonly used to generate stable, lipid-incorporated labeling of biological membranes and membrane-derived materials, providing a convenient route to visualize membrane structure and dynamics with standard fluorescence microscopy and imaging instrumentation.
1. Membrane Labeling Studies
ATTO 532 DMPE is frequently used in fluorescence microscopy and imaging experiments to label cell membranes and model lipid membranes by incorporating the DMPE lipid into lipid bilayers. Researchers use it for visualizing membrane morphology, tracking membrane-associated processes, and comparing membrane organization across experimental conditions. The ATTO 532 emission is leveraged for multicolor imaging workflows where robust red/orange channel signal is required, and the lipid anchor supports membrane retention during wash steps and imaging buffers.
2. Liposome And Nanoparticle Tracing
ATTO 532 DMPE is widely applied for labeling liposomes and lipid-based nanoparticles to enable fluorescence tracing of particle uptake, trafficking, and surface interactions in cell culture and biomaterials studies. Because the dye is delivered via a phospholipid handle, it integrates into lipid-containing carriers rather than relying on nonspecific adsorption of a free dye. This makes it useful for preparing labeled delivery vehicles for imaging-based characterization of binding and internalization, as well as for monitoring particle stability in assay media.
3. Flow Cytometry Membrane Dyes
ATTO 532 DMPE is used as a membrane-associated fluorescence reagent for flow cytometry workflows where membrane labeling provides a readout of cell-associated signal. In typical experimental setups, it helps generate a stable fluorescence signal tied to the cell surface or membrane fraction, supporting comparative analysis of populations under different labeling or treatment conditions. Researchers also use it when they need a dye that behaves as a lipid-incorporating probe rather than a purely reactive conjugate, simplifying staining workflows for membrane-focused assays.
4. FRET Pair Construction
ATTO 532 DMPE is also used in fluorescence resonance energy transfer (FRET) assay development when ATTO 532 is selected as the donor fluorophore within lipid environments. In membrane-mimetic systems and lipid-based assay formats, the DMPE anchor supports consistent dye placement in bilayers, which is important for constructing donor-acceptor architectures used to monitor proximity-dependent energy transfer. Teams developing membrane FRET reporters often incorporate ATTO 532 DMPE alongside complementary acceptor dyes to build lipid-integrated sensing constructs for imaging and plate-based fluorescence measurements.
