Product Introduction
ATTO 514 is a new hydrophilic fluorescent label with excellent water solubility. The dye exhibits strong absorption, high fluorescence quantum yield and exceptional thermal and photo-stability. Thus ATTO 514 is highly suitable for single-molecule detection applications and high-resolution microscopy such as PALM, dSTORM, STED etc. Additionally the dye highly qualifies to be applied in flow cytometry (FACS), fluorescence in-situ hybridization (FISH) and many more. The fluorescence is excited most efficiently in the range 510-535 nm. A suitable source of excitation is the 514 nm line of the Argon-Ion laser.find more information here

Application
ATTO 514 is a fluorescent dye designed for robust labeling workflows, typically used to generate bright, spectrally distinct conjugates for fluorescence imaging and quantitative fluorescence assays. Its reactive handle enables attachment to biomolecules and polymers, supporting downstream probe construction, conjugate tracking, and signal readout on common fluorescence instrumentation.
1. Fluorescence Labeling Conjugates
ATTO 514 is frequently used by chemistry and biology teams to prepare fluorescently labeled proteins, peptides, and other research biomolecules for imaging and binding studies. In these workflows, the dye's attachment chemistry is leveraged to generate stable conjugates that can be tracked during assay development, purification monitoring, and comparative studies of labeled vs. unlabeled reagents. Researchers also use ATTO 514-labeled conjugates as fluorescent reporters in multi-step bioconjugation pipelines, including conjugate characterization by fluorescence-based analytics.
2. Fluorescence Microscopy Imaging
ATTO 514 supports fluorescence microscopy experiments where a far-red/green-red spectral channel is needed for cellular and molecular visualization. Imaging groups use ATTO 514-labeled antibodies, affinity reagents, or polymeric materials to visualize binding patterns, localization trends, and spatial distribution in fixed-cell or fixed-sample formats, as well as in labeled biomaterial studies where fluorescence tracking is required. The dye's brightness and practical labeling utility make it a common choice for constructing imaging reagents for confocal and widefield fluorescence microscopy.
3. Flow Cytometry Fluorescent Tracing
ATTO 514 is used in flow cytometry workflows to label targets for fluorescence-based population analysis and reagent tracing. In typical assay development, ATTO 514 conjugates are incorporated into staining panels to distinguish labeled samples from controls and to quantify relative fluorescence intensity across cell populations. Researchers also apply ATTO 514 labeling to monitor labeling efficiency and to track conjugate uptake or association in experimental systems where flow readout is the primary measurement platform.
4. Fluorescence Bioassay Development
ATTO 514 is widely adopted in fluorescence assay development as a reporter dye for constructing labeled assay reagents, including fluorescent substrates, labeled binding components, and fluorescence readout standards. Assay developers use ATTO 514-labeled constructs to enable plate-based fluorescence measurements of binding events, reagent performance, and workflow consistency during optimization. The dye's conjugation versatility supports rapid iteration of assay reagent formats for research-grade screening and method development.
