Product Introduction
ATTO 490LS is a new fluorescent label featuring an extraordinary large Stokes-Shift of 165 nm. Thus the emission spectrum is almost completely separated from its absorption spectrum, making the dye highly suitable for multiplexing experiments, in particular in combination with ATTO 488 and ATTO 514. ATTO 490LS is very hydrophillic and shows excellent water solubility. The dye exhibits a relatively high fluorescence quantum yield, which is only slightly reduced after conjugation to biomolecules, e.g. proteins, even at high degrees of labeling (DOL). ATTO 490LS is an anionic dye. After conjugation to a substrate the dye carries a net electrical charge of -1.find more information here

Application
ATTO 490LS NHS ester is a reactive, NHS-ester-activated ATTO 490LS fluorophore designed for efficient covalent fluorescent labeling of primary amine-containing biomolecules. Its bright visible emission supports fluorescence microscopy and flow cytometry workflows where robust dye conjugates are required, and the NHS ester enables straightforward post-conjugation labeling of proteins, peptides, and antibody constructs.
1. Protein And Antibody Labeling
ATTO 490LS NHS ester is widely used for preparing fluorescently labeled proteins and antibody conjugates for cellular and biochemical assays. Researchers typically apply it to label lysine-rich targets such as enzymes, serum proteins, and affinity reagents to generate imaging-compatible conjugates for binding studies, target localization experiments, and fluorescence-based readouts. The NHS-ester functionality supports stable covalent attachment under standard labeling conditions, producing dye-protein conjugates that can be used in microscopy and plate-based fluorescence measurements.
2. Fluorescence Microscopy Imaging
ATTO 490LS NHS ester supports fluorescence microscopy applications that require a visible-spectrum dye for cellular imaging and colocalization experiments. By labeling antibodies, lectins, or other amine-bearing probes, investigators generate fluorescent staining reagents for fixed-cell workflows and for structured imaging experiments where consistent labeling density is important. The ATTO 490LS emission is commonly leveraged in multicolor imaging panels by pairing with spectrally distinct fluorophores, enabling spatial mapping of biomolecular distributions on cells and in labeled biomaterials.
3. Flow Cytometry Dye Conjugates
ATTO 490LS NHS ester is used to generate fluorescent conjugates for flow cytometry, especially when amine-reactive labeling of antibodies or protein binders is required. Flow cytometry users rely on covalent dye attachment to produce stable antibody-fluorophore conjugates for detecting surface markers, intracellular targets after permeabilization, or labeling of cell-associated biomolecules in suspension. The dye's visible emission region makes it practical for instrument configurations that include blue/green excitation and corresponding emission detection channels.
4. Fluorescent Labeling Of Peptides
ATTO 490LS NHS ester enables fluorescent labeling of peptides and peptide-based affinity reagents for imaging and assay development. In chemical biology workflows, researchers label amine-containing peptides to create fluorescent tracers for binding and uptake studies, as well as for monitoring peptide interactions in solution. Covalent NHS-ester labeling helps maintain probe integrity during washing steps and downstream assay handling, supporting reproducible fluorescence readouts in microscopy and fluorescence assays.
