Product Introduction
ATTO 488 is a labeling dye with high molecular absorption (90,000) and quantum yield (0.80) as well as sufficient Stokes shift between excitation and emission maximum. It is optimized for excitation with an argon laser, and is characterized by high photostability.ATTO-Dye Labeled PhospholipidsSigma-Aldrich offers a variety of glycero-phospholipids carrying one or two fatty acid groups (lipophilic groups) and a phosphate ester residue (hydrophilic group). They are labeled at the hydrophilic head group. After incorporation of the phospholipid into a membrane the fluorophore is located at the water/lipid interface of the membrane. We currently provide 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE), 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), palmitoyl-sn-glycero-phosphoethanolamine (PPE), and 1,2-dimyristoyl-snglycero-3-phospho-ethanolamine (DMPE) labeled with ATTO-dyes.find more information here

Application
ATTO 488 DOPE is a fluorescent lipid probe designed for membrane labeling, built on a DOPE (dioleoyl phosphatidylethanolamine) scaffold bearing an ATTO 488 dye for green fluorescence imaging. Its amphiphilic lipid character enables incorporation into lipid bilayers, supporting visualization of membranes and membrane-associated processes by fluorescence microscopy and related optical readouts.
1. Membrane Labeling Microscopy
ATTO 488 DOPE is widely used by cell imaging and membrane biophysics groups to fluorescently tag lipid bilayers for confocal and widefield fluorescence microscopy. Researchers incorporate the DOPE-lipid into model membranes or cellular membranes to track membrane organization, membrane trafficking intermediates, and lipid mixing events. The ATTO 488 fluorophore provides a bright green channel suitable for multi-color imaging workflows when paired with spectrally distinct dyes, enabling spatial mapping of labeled membrane domains during time-lapse experiments.
2. Liposome And Nanoparticle Tracing
ATTO 488 DOPE supports fluorescent labeling of liposomes and lipid-based nanoparticles for studies in delivery research and colloidal characterization. By embedding the dye-labeled lipid into the particle membrane, investigators can monitor particle integrity, membrane association, and uptake-related dynamics using fluorescence microscopy or plate-based fluorescence assays. This use is particularly common in formulation screening and in vitro characterization where membrane-associated fluorescence provides a practical readout for comparing different lipid compositions and preparation conditions.
3. Flow Cytometry Lipid Staining
ATTO 488 DOPE is used by fluorescence cytometry users to stain membrane-containing particles and cells for flow cytometry readouts based on membrane fluorescence intensity. In membrane-labeling workflows, the DOPE lipid integrates into lipid structures, allowing quantification of fluorescence signal across populations and enabling gating strategies for labeled versus unlabeled samples. This approach is frequently applied in assays that compare membrane composition, membrane binding of lipid-associated reagents, or relative labeling levels across experimental conditions.
4. FRET Pair Membrane Assays
ATTO 488 DOPE is also employed in FRET-based membrane studies where ATTO 488 serves as a donor fluorophore in lipid environments. Researchers construct donor-labeled membrane systems by incorporating ATTO 488 DOPE into lipid mixtures and pairing it with an appropriate acceptor dye to report on proximity-dependent energy transfer within membranes. Such membrane FRET assays are commonly used to investigate lipid mixing, membrane fusion-related events, and nanoscale organization using fluorescence intensity or acceptor emission readouts.
