Product Introduction
ATTO 430LS is a new fluorescent label featuring an extraordinary large Stokes-Shift of 114 nm. Thus its emission spectrum is almost completely separated from its absorption spectrum, making the dye highly suitable for multiplexing experiments. ATTO 430LS is very hydrophilic and shows excellent water solubility. The dye exhibits a high fluorescence quantum yield, which is only slightly reduced after conjugation to biomolecules, e.g. proteins, even at high degrees of labeling (DOL).ATTO 430LS is an anionic dye. After conjugation to a substrate the dye carries a net electrical charge of -1. The fluorescence is excited most efficiently in the range 400-460 nm.find more information here

Application
ATTO 430LS NHS ester is an amine-reactive fluorescent dye designed for efficient covalent labeling of proteins and peptides via NHS ester chemistry. This dye is typically used in visible-range fluorescence workflows where bright, photostable signal is required for microscopy, labeling controls, and quantitative fluorescence readouts. Because it reacts with primary amines, it is widely adopted for preparing fluorescent conjugates for downstream bioimaging and assay development.
1. Protein And Antibody Labeling
ATTO 430LS NHS ester is used by protein chemistry and chemical biology groups to generate fluorescently labeled proteins, including antibodies, antibody fragments, and enzyme conjugates for fluorescence-based characterization. The NHS ester functionality enables straightforward attachment to lysine residues and N-termini, producing stable covalent fluorescent conjugates that are commonly used as staining reagents, binding probes, and reference standards in imaging experiments. Researchers often prepare these conjugates for microscopy studies and for comparing labeling density across experimental conditions, including workflow development for immunofluorescence-like assays and fluorescence binding experiments.
2. Fluorescence Microscopy Staining
ATTO 430LS NHS ester supports fluorescence microscopy workflows where covalent labeling is preferred over noncovalent dye association. Cell biology and microscopy teams use dye-labeled proteins and peptides to visualize biomolecular localization, track binding interactions on cell surfaces, and generate fluorescent tracers for fixed-sample imaging. In these applications, the dye's visible excitation/emission characteristics align well with standard fluorescence microscope filter sets, enabling robust imaging of labeled targets while maintaining the conjugate stability required for washing steps and multi-step staining protocols.
3. Flow Cytometry Labeling Reagents
ATTO 430LS NHS ester is frequently employed to prepare fluorescent labeling reagents for flow cytometry, particularly when covalent dye attachment to targeting biomolecules is desired. Immunology and cell analysis laboratories use NHS-labeled proteins to quantify binding to cell-associated epitopes, to monitor receptor engagement in binding assays, and to create consistent fluorescence standards for instrument setup and gating controls. Covalent conjugation helps reduce dye loss during sample handling, supporting reproducible signal across runs when labeled antibodies or protein ligands are used as flow-compatible staining reagents.
4. Fluorescent Assay Reagent Development
ATTO 430LS NHS ester is also used in fluorescence assay development to construct labeled reagents for binding studies, kinetic monitoring, and assay calibration. Assay developers label proteins, peptides, or other primary-amine-containing components to create fluorescent readouts that can be measured in plate readers or fluorescence imaging systems. The dye provides a practical handle for building assay-specific conjugates, including fluorescent tracking reagents for workflow optimization, reagent qualification, and comparative studies where consistent covalent labeling is important for interpreting fluorescence signals.
