Product Introduction
TAMRA Sulfo-NHS ester is a reactive form of bright yellow dye that used to generate a stable fluorescence signal in bioimaging. The maxima of Ex/Em values are at 554/583 nm, similar to that of DyLight 549, ATTO 550 and Cy 3. TAMRA might be excited using 543 or 546 nm laser line and displays good optical property. Sulfo-NHS esters have higher water solubility than NHS esters, thus they do not need organic co-solvent and readily react with amine-modified oligonucleotides or amino groups of proteins, i.e. the ε-amino groups of lysine or the amine terminus of nucleotides to form a stable amide bond between dye and the biomolecule. We offer TAMRA Sulfo-NHS ester for labeling of antibodies, peptides, proteins and oligonucleotide labeling and automated DNA sequencing applications.

Application
TAMRA Sulfo-NHS ester is a sulfonated, NHS-ester activated derivative of the TAMRA fluorophore designed for efficient amine-reactive fluorescent labeling in aqueous buffers. Its charged sulfonate group supports water compatibility for bioconjugation workflows, including labeling of proteins, peptides, and other primary-amine containing biomolecules. The TAMRA emission is commonly used in fluorescence microscopy and fluorescence-based assays where red/orange detection channels are available.
1. Protein And Antibody Labeling
TAMRA Sulfo-NHS ester is widely used to generate fluorescently labeled proteins and antibodies for binding studies, immunoassays, and fluorescence imaging experiments. Researchers typically employ the NHS ester to covalently tag lysine residues on target biomolecules, producing stable conjugates for tracking binding, monitoring reagent distribution, and constructing fluorescent detection reagents. The sulfonate functionality helps keep the dye conjugates water-compatible, which is advantageous when preparing labeling stocks for buffer-based assay formats and microscopy sample preparation.
2. Peptide And Biomolecule Conjugation
TAMRA Sulfo-NHS ester supports fluorescent labeling of peptides and other amine-bearing biomolecules used as tracers in chemical biology and molecular imaging workflows. In practice, it is used to prepare TAMRA-tagged conjugates for studying biomolecular interactions, validating conjugation strategies, and generating fluorescent standards for method development. Because the reagent reacts with primary amines, it is commonly selected when a straightforward, covalent labeling step is needed to create a robust fluorescent probe without relying on noncovalent dye adsorption.
3. Fluorescence Microscopy Staining Probes
TAMRA Sulfo-NHS ester is used to prepare red/orange fluorescent labeling reagents for microscopy workflows where TAMRA emission can be resolved from other channels. Typical use cases include labeling biomolecule components that will be visualized in fixed-cell or fixed-tissue imaging experiments, as well as generating fluorescent conjugates for co-localization studies with other fluorophores. The sulfonated dye format helps maintain aqueous handling during conjugate preparation and downstream staining steps, supporting consistent sample preparation for imaging and quantitative fluorescence readouts.
4. Flow Cytometry Labeling Reagents
TAMRA Sulfo-NHS ester is frequently incorporated into fluorescent labeling reagent development for flow cytometry, particularly when a TAMRA-compatible detection channel is available. Researchers use the NHS ester to covalently attach TAMRA to proteins used as staining reagents, enabling fluorescence-based analysis of labeled biomolecules in cell-associated assays. Water-compatible conjugate preparation is helpful for generating reproducible staining reagents for assay development and instrument method optimization.
