Product Introduction
ATTO 700 belongs to a new generation of fluorescent labels. The dye is designed for application in the area of life science, e.g. labeling of DNA, RNA or proteins. Characteristic features of the label are strong absorption, high fluorescence quantum yield, excellent thermal and photo-stability, very good water solubility and very little triplet formation. ATTO 700 is a zwitterionic dye with a net electrical charge of zero. The fluorescence is efficiently quenched by electron donors like guanine, tryptophan, etcPhalloidin is a fungal toxin isolated from the poisonous mushroom Amanita phalloides. Its toxicity is attributed to the ability to bind F actin in liver and muscle cells. As a result of binding phalloidin, actin filaments become strongly stabilized. Phalloidin has been found to bind only to polymeric and oligomeric forms of actin, and not to monomeric actin. The dissociation constant of the actin-phalloidin complex has been determined to be on the order of 3 x 10-8. Phalloidin differs from amanitin in rapidity of action; at high dose levels, death of mice or rats occurs within 1 or 2 hours. Fluorescent conjugates of phalloidin are used to label actin filaments for histological applications. Some structural features of phalloidin are required for the binding to actin. However, the side chain of amino acid 7 (g-d-dihydroxyleucine) is accessible for chemical modifications without appreciable loss of affinity for actin.find more information here

Application
Phalloidin-ATTO 700 is a fluorescent phalloidin conjugate designed for actin filament visualization. The phalloidin scaffold provides strong binding to filamentous actin (F-actin), while the ATTO 700 dye enables bright far-red fluorescence for microscopy-based localization studies. This reagent is widely used in cell biology workflows where robust actin staining and clear separation from green spectral channels are required.
1. F-Actin Fluorescence Microscopy
Phalloidin-ATTO 700 is used by cell biology and microscopy laboratories to label filamentous actin in fixed cells for spatial mapping of cytoskeletal organization. Researchers incorporate the conjugate into immunofluorescence-style staining workflows to visualize stress fibers, cortical actin, and actin-rich structures, often alongside other fluorophores in multicolor imaging panels. The far-red emission of ATTO 700 helps reduce spectral overlap when combined with common green/yellow dyes, supporting cleaner channel separation in widefield fluorescence and confocal microscopy.
2. Cytoskeleton Co-Localization Studies
Phalloidin-ATTO 700 supports quantitative and qualitative co-localization experiments where actin filaments are compared with proteins, signaling complexes, or membrane-associated markers. Chemical biology groups and imaging core facilities use the reagent to generate consistent actin reference signals for downstream image analysis, including colocalization metrics and segmentation-based quantification of actin architecture. Because phalloidin binding targets F-actin rather than monomeric actin, the stain is commonly selected when the experimental goal is filament-specific structural readouts.
3. Fixed-Cell Actin Staining Panels
Phalloidin-ATTO 700 is frequently selected for standardized fixed-cell staining panels in phenotypic assays and microscopy screening workflows that require reproducible cytoskeletal labeling. Researchers use the conjugate to prepare actin-positive reference samples for protocol development, antibody panel optimization, and imaging parameter tuning (exposure, laser power, and filter sets). The ATTO 700 far-red signal is particularly useful when building multichannel experiments that include multiple fluorescent labels, enabling actin to be assigned to a distinct spectral window for robust imaging readouts.
4. High-Content Imaging Readouts
Phalloidin-ATTO 700 is applied in high-content imaging and automated microscopy workflows to generate actin filament features used for morphology scoring and cytoskeletal phenotype classification. Screening teams in academic and industrial research environments use the reagent to produce consistent actin staining across plates, supporting automated segmentation of filamentous structures and extraction of texture and shape descriptors. Its far-red fluorescence is commonly leveraged to streamline assay design in multiplexed imaging formats where actin labeling must remain distinguishable from other fluorophores in the same well.
