Background

pH is closely related to various life activities of living organisms. As one of the main parameters of cell metabolism, intracellular pH plays a vital role in many physiological and pathological processes such as cell growth, cell proliferation, cell apoptosis, cell endocytosis, ion transfer, signal transmission and multi-drug resistance. The subtle pH changes in cells are closely related to cell functions. Abnormal pH changes may cause cell dysfunction and cause a variety of diseases, such as cancer, stroke, and Alzheimer's disease. Therefore, the detection of intracellular pH changes is of high significance for studying the physiological and pathological processes of living organisms, and is helpful for the treatment of related diseases. At present, the methods for detecting intracellular pH mainly include microelectrode method, nuclear magnetic resonance method, absorption spectroscopy and fluorescence method. Fluorescence method has the advantages of simple operation, high sensitivity, good selectivity, real-time detection and little damage to organisms. The key to the fluorescence method is the fluorescent probes.

pH-sensitive fluorescent probes/ indicators

pH-sensitive fluorescent probes/indicators are a class of substances whose fluorescent properties change with changes in pH. The qualitative measurement of pH can be achieved by the increase or decrease of the fluorescence intensity of the pH indicator in a specific pH range. In addition, as the pH changes, the ratio of the probe molecule's spectra changes can achieve a quantitative measurement of pH. After the pH indicator enters the cell, it can be observed with a fluorescence microscope to detect the dynamic distribution and regional changes of the pH in the cell in real time.

pH indicators are widely used to detect pH levels in cells. pH indicators are widely used in the processes of cell proliferation and apoptosis, drug resistance, and cell chemotaxis with changes in intracellular pH.
