Background

ATTO is the most common single molecule fluorescent dye, which can be used as fluorescent labeling and molecular probe for a series of biological molecules such as protein and nucleic acid. Its spectrum covers the range from ultraviolet to near-infrared light. It is the most complete fluorescent label. Compared with other dyes, it has excellent light stability and brightness in the red spectral region.

Characteristics of ATTO

The ATTO series of dyes have a spectral range of 390nm to 740nm and can be excited by ordinary light sources. Compared with other dyes, ATTO dyes exhibit excellent photostability and brightness, especially in the red spectral region. For example, in aqueous solution, ATTO 647N produces twice as much fluorescence as Cy5. Even when exposed to ozone, the new dye ATTO remains highly reactive. In addition, ATTO dyes have the characteristics of strong fluorescence signal, large molar absorption coefficient, low excitation or low emission overlap, which make ATTO an ideal dye for multiplexing technology.

Application of ATTO

ATTO is named after their respective excitation wavelengths. ATTO dyes are usually coumarin, rhodamine, azine and other derivatives. Compared with dyes that are used for a long time and have a wide range of applications, ATTO dyes are more stable under long irradiation. Common ATTO dyes are ATTO 465, ATTO 488, ATTO 647, etc. All can be linked to carboxy, nhs-ester, maleimide, hydrazide, biotin, phalloidin, amine, azide, iodoacetamide, alkyne, cadaverine and other groups.

  • ATTO fluorescent dyes, due to their high sensitivity and convenient operation, have gradually replaced radioisotopes as detection markers. They are widely used in fluorescent immunity, fluorescent probes, cell staining, etc., including specific DNA staining for chromosome analysis, cell cycle, cell apoptosis and other related research. In addition, many nucleic acid dyes are very useful re staining agents in multicolor dyeing system, which can be used as background control to mark the nucleus, so as to make the spatial relationship of intracellular structure clear at a glance.

  • Fluorescent labeled monoclonal antibody technology has expanded unlimited application space for flow cytometry in the study of cell membrane, well-known species functional antigen, tumor gene protein and other fields. ATTO fluorescent dyes can be covalently bound to monoclonal antibody through protein cross linker for fluorescence labeling.

  • Nucleic Acids Detection
  • After the nucleus is stained with nucleic acid ATTO dye, the fluorescence intensity emitted by the cell can be measured quantitatively, the content of DNA and RNA in the nucleus can be determined, and the cell cycle and cell proliferation can be analyzed.
