Golgi Fluorescent Probes are cell-penetrating probes. They can selectively stain the Golgi complex for lipid metabolism and transport studies. At present, there are two commercialized probes for locating the Golgi apparatus, namely the Golgi red fluorescent probe and the Golgi green fluorescent probe. The maximum excitation and emission wavelengths of the Golgi green fluorescent probes are 505 nm and 511 nm, respectively. The maximum excitation and emission wavelengths of Golgi red fluorescent probes are 589 nm and 617 nm, respectively. Golgi Fluorescent Probes are stored in -20 ℃. They must avoid repeated freezing and thawing.
Characteristics of Golgi fluorescent probes
Golgi fluorescent probes have concentration-dependent optical properties. The stock solution of Golgi fluorescent probes was dissolved in DMSO at a concentration of 1 mM, and the recommended working concentration is 5 μM. Golgi red and green fluorescent probes are both types of sphingolipid fluorescent probes and can be used for Golgi-specific fluorescent staining of living cells. Golgi fluorescent probes inevitably have quenching problems. Please avoid light as much as possible during storage and operation to slow down the quenching of fluorescence.
Application of Golgi fluorescent probes
Golgi fluorescent probes are often used for lipid transport and metabolism studies in living cells. Compared with the traditional probe of the same type, Golgi fluorescent probes exhibit higher molar absorption coefficient and photon yield, and stronger photostability. In addition to the above applications, the probes can also be used to measure the rate of lipid synthesis in Schwann cells, and to label cell outlines for observation of morphological movements under confocal microscopy. Among them, Golgi red fluorescent probes can be used for Golgi fluorescent labeling of living cells, but it is not suitable for labeling of fixed cells.
