    Trans cyclooctene (TCO) fluorescent probe mainly relies on the click reaction of trans cyclooctene (TCO) for specific labeling. After bio-orthogonal reaction with tetrazine (TZ), a series of fluorescent molecules modified on dienes were released through a series of reaction processes. Among them, the click release reaction between TCO and TZ has the advantages of fast reaction, no catalysis and non-toxic by-products (N2 and CO2). It has great application potential in the fields of prodrug preparation, drug administration and antibody drug coupling.

    Characteristics of TCO

    Trans cyclooctene (TCO) can be used for strain promoted copper free click chemical cycloaddition reaction with 1,2,4,5-tetrazine. The cyclooctene can react with tetrazine functionalized compounds or biomolecules without the presence of catalysts, and can produce stable covalent bonds. Diels-alder cycloaddition reaction between trans cyclooctene and tetrazine is the fastest biocompatible bonding technology reported, and has many applications in biomarkers and imaging. With the help of efficient TCO and TZ biological orthogonal reaction, the technology can achieve ultra-fast fluorescence quenching in 1 second. Within 1h, imaging analysis of 20-30 markers can theoretically be achieved through multiple rounds of markers.

    Application of TCO

    Biorthogonal reaction has become an effective tool to study the mechanism and function of bioactive molecules such as peptides, proteins and antibodies in biological systems. TZ functionalized molecules and TCO functionalized molecules form stable conjugates through partial reaction of dihydropyrazine. Because TCO may have very fast cyclo addition kinetics as a dienophilic reagent, this anti electron demand cyclo addition reaction has become common. Dyes containing TZ and TCO can be used to explore various biological systems that can use click reaction. For example, fluorescent labels and quenchers are constructed based on TCO and TZ respectively for single-cell fluorescence imaging and rapid quenching of fluorescence signals. Cy5-TCO has been used to label biomolecules for fluorescence biochemical analysis such as fluorescence imaging, and is widely used to label peptides, proteins and oligonucleotides.
